....that despite the Bible's encouragement to eat, drink, enjoy one's life with one's wife, and so on, the notion that holiness and joy/happiness are somehow opposed persists...

There's always plenty of legalism to go around, unfortunately. When we spend more time worrying about what we can and can't do on Sunday than actually enjoying God's good gifts, it's pretty clear we're growing neither in holiness nor happiness.
